Understanding features of the ASP.NET framework is absolutely necessary.  We write all ASP.NET code in C#.  Understanding fundamentals of object oriented programming, class design, recursion, interfaces, enumerations, and conventions is necessary.  Desktop application programming experience is a plus, though, not a requirement.  Experience with multithreading, reflection, interop, is a plus, though, not a requirement.  Microsoft certifications are a plus, though, not a requirement.
